Uncertain significance for EGFR-related lung cancer — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005228.5(EGFR):c.110A>G (p.Lys37Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces lysine, which is basic and polar, with arginine, which is basic and polar, at codon 37 of the EGFR protein (p.Lys37Arg). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with EGFR-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Benign". The arginine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. Experimental studies have shown that this missense change affects EGFR function (PMID: 26244656). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
