Uncertain significance for Hereditary spastic paraplegia 39 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001166114.2(PNPLA6):c.3979G>T (p.Gly1327Trp), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with PNPLA6-related disease. This variant is not present in population databases (ExAC no frequency). This sequence change replaces glycine with tryptophan at codon 1289 of the PNPLA6 protein (p.Gly1289Trp). The glycine residue is moderately conserved and there is a large physicochemical difference between glycine and tryptophan. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
