This variant lies in the SLC2A1 gene (transcript NM_006516.4) at coding-DNA position 1167, where C is replaced by A; at the protein level this means replaces phenylalanine at residue 389 with leucine — a missense variant. Submitter rationale: This sequence change replaces phenylalanine, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 389 of the SLC2A1 protein (p.Phe389Leu). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with glucose transporter type 1 deficiency syndrome (PMID: 32134591). In at least one individual the variant was observed to be de novo. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant.
