This variant lies in the POLD1 gene (transcript NM_002691.4) at coding-DNA position 2218, where A is replaced by G; at the protein level this means replaces threonine at residue 740 with alanine — a missense variant. Submitter rationale: This sequence change replaces threonine, which is neutral and polar, with alanine, which is neutral and non-polar, at codon 740 of the POLD1 protein (p.Thr740Ala). This variant is present in population databases (rs781629170, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with POLD1-related conditions. ClinVar contains an entry for this variant (Variation ID: 469243).
