Uncertain significance for Hypogonadotropic hypogonadism 1 with or without anosmia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000216.4(ANOS1):c.815G>A (p.Arg272Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 272 of the ANOS1 protein (p.Arg272Gln). This variant is present in population databases (rs749764438, gnomAD 0.008%), including at least one homozygous and/or hemizygous individual. This variant has not been reported in the literature in individuals affected with ANOS1-related conditions. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt ANOS1 protein function with a positive predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
