Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_016616.5(NME8):c.1648G>T (p.Asp550Tyr), citing Ambry Variant Classification Scheme 2023: The p.D550Y variant (also known as c.1648G>T), located in coding exon 15 of the NME8 gene, results from a G to T substitution at nucleotide position 1648. The aspartic acid at codon 550 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
