Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001330311.2(DVL1):c.1993G>T (p.Glu665Ter), citing LabCorp Variant Classification Summary - May 2015: Variant summary: DVL1 c.1918G>T (p.Glu640X) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, however current evidence is not sufficient to establish loss of function as a mechanism for disease. The variant was absent in 157316 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1918G>T in individuals affected with DVL1-related conditions and no experimental evidence demonstrating its impact on protein function have been reported. While numerous frameshift variants have been reported to generate nascent stop codons near the location of the present variant, the literature has not yet defined a result for nonsense variants expected to escape nonsense mediate decay (PMID: 26924530, 25817016, 25817014).
