Pathogenic for Polymicrogyria with or without vascular-type Ehlers-Danlos syndrome — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000090.4(COL3A1):c.3712del (p.Leu1238fs), citing LabCorp Variant Classification Summary - May 2015: Variant summary: COL3A1 c.3712delC (p.Leu1238SerfsX8) results in a premature termination codon, predicted to cause absence of the protein due to nonsense mediated decay, which is a commonly known mechanism for disease. The variant was absent in 251374 control chromosomes. To our knowledge, no occurrence of c.3712delC in individuals affected with COL3A1-related conditions and no experimental evidence demonstrating its impact on protein function have been reported. No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as pathogenic.
