Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001042681.2(RERE):c.2762C>T (p.Ala921Val), citing LabCorp Variant Classification Summary - May 2015: Variant summary: RERE c.2762C>T (p.Ala921Val) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant allele was found at a frequency of 8.8e-06 in 1592820 control chromosomes. This frequency is not significantly higher than the estimated maximum expected for disease-causing variants in RERE, allowing no clear conclusions about variant significance, although occurrence in multiple carriers suggests that this variant is likely not associated with a high penetrance, severe, early onset disease phenotype in heterozygous state. To our knowledge, no occurrence of c.2762C>T in individuals affected with RERE-related conditions and no experimental evidence demonstrating its impact on protein function have been reported. No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as uncertain significance.
