A presumed nomenclature of c.(3930+1_3931-1)_(*110_?)dup has been designated for the purposes of this classification. The exact breakpoint at the 3' end of this variant is unknown, therefore this duplication may extend downstream of the annotated region of the gene. As it duplicates the termination codon, its effect on the encoded protein is unknown. A duplication of exons 19-29 that also involves part of the CKMT1B gene was found at a frequency of 0.014 in gnomAD, including one homozygote; however the current variant (duplication of exons 20-29 in STRC) was absent in 21592 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.(3930+1_3931-1)_(*110_?)dup in individuals affected with STRC-related conditions and no experimental evidence demonstrating its impact on protein function have been reported.
