Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NC_000012.11:g.(?_56078351)_(56101689_?)dup, citing LabCorp Variant Classification Summary - May 2015: Variant summary: The variant involves the duplication of exons 1-25 in the ITGA7 gene. A presumed nomenclature of c.(?_-223)_(*491_?)dup has been designated for the purposes of this classification. This duplication includes the entire coding sequence of the gene. As exact breakpoints are unknown, it may extend beyond the annotated region of the gene, to include other flanking genes. The duplication of the ITGA7 gene (in isolation) was absent in 120780 control chromosomes in the gnomAD database (Structural Variants v4.1 dataset). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.(?_-223)_(*491_?)dup in individuals affected with ITGA7-related conditions and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 832932). Based on the evidence outlined above, the variant was classified as uncertain significance.
