This variant lies in the C1R gene (transcript NM_001733.7) at coding-DNA position 446, where G is replaced by A; at the protein level this means replaces arginine at residue 149 with glutamine — a missense variant. Submitter rationale: Variant summary: C1R c.446G>A (p.Arg149Gln) results in a conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be tolerated. The variant allele was found at a frequency of 3.8e-05 in 780824 control chromosomes, predominantly at a frequency of 5.3e-05 within the Non-Finnish European subpopulation in the gnomAD database. The observed variant frequency within Non-Finnish European control individuals in the gnomAD database exceeds the estimated maximal expected allele frequency for disease-causing variants in C1R. c.446G>A has been observed in the heterozygous state in an individual affected with Gorham-Stout Disease (example: Rudilla_2019). This report does not provide unequivocal conclusions about association of the variant with Ehlers-Danlos syndrome, periodontal type 1. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
