Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 251304 control chromosomes. To our knowledge, no occurrence of c.545G>C in individuals affected with STAR-related conditions and no experimental evidence demonstrating its impact on protein function have been reported. Different variant(s) affecting the same codon has been classified as pathogenic by our lab (p.Arg182Cys, p.Arg182Leu), supporting the critical relevance of codon 182 to STAR protein function. No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as VUS-possibly pathogenic.
