This variant lies in the ELP1 gene (transcript NM_003640.5) at 5 bases into the intron immediately after coding-DNA position 303, where G is replaced by A. Submitter rationale: Variant summary: ELP1 c.303+5G>A alters a nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Several computational tools predict a significant impact on normal splicing: Three predict the variant weakens a 5' donor site. One predicts the variant has no significant impact on splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 6.2e-07 in 1613964 control chromosomes (gnomAD v4). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance.
