Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_005548.3(KARS1):c.3G>A (p.Met1Ile), citing LabCorp Variant Classification Summary - May 2015: Variant summary: KARS1 c.-94G>A is located in the untranslated mRNA region upstream of the initiation codon. This variant, also annotated as NM_005548.3 c.1A>G (p.Met1? aka p.Met1Val) results in an initiation codon variant within an alternate transcript; however currently, loss-of-function is not a known mechanism of disease for this transcript (see e.g. PMID: 29615062). The variant was absent in 249060 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.-94G>A in individuals affected with KARS1-related conditions and no experimental evidence demonstrating its impact on protein function have been reported. No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as uncertain significance.
