Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001206744.2(TPO):c.962C>A (p.Thr321Asn), citing LabCorp Variant Classification Summary - May 2015: Variant summary: TPO c.962C>A (p.Thr321Asn) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 238444 control chromosomes. c.962C>A has been observed in individuals in one family affected with Deficiency Of Iodide Peroxidase (Furman_2021). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
