Likely pathogenic for Mucopolysaccharidosis, MPS-IV-B — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000404.4(GLB1):c.485T>C (p.Leu162Ser), citing LabCorp Variant Classification Summary - May 2015: Variant summary: GLB1 c.485T>C (p.Leu162Ser) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 203182 control chromosomes (gnomAD). c.485T>C has been observed in an individual affected with Mucopolysaccharidosis Type IVB (Morquio Syndrome B)(Santamaria_2007a). These data indicate that the variant may be associated with disease. At least one publication reports experimental evidence evaluating an impact on protein function, finding that the variant results in no residual enzymatic activity (Santamaria_2007b). The following publications have been ascertained in the context of this evaluation (PMID: 17309651, 17664528). No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as likely pathogenic.
