Submitter rationale: ABCB4 p.Cys717Ser (c.2149T>A) is a missense variant that changes the amino acid at residue 717 from Cysteine to Serine. This variant has been observed in at least one proband with an ABCB4-related disorder (PMID:33757843;31115781). The variant was found to segregate with disease in at least one affected family (PMID:31115781). It is absent or not present at a significant frequency in gnomAD. In silico models predict that this variant is possibly or probably damaging. In conclusion, we classify ABCB4 p.Cys717Ser (c.2149T>A) as a variant of uncertain significance.
