Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000314.8(PTEN):c.172G>A (p.Asp58Asn), citing Ambry Variant Classification Scheme 2023: The p.D58N variant (also known as c.172G>A), located in coding exon 3 of the PTEN gene, results from a G to A substitution at nucleotide position 172. The aspartic acid at codon 58 is replaced by asparagine, an amino acid with highly similar properties. In a massively parallel functional assay using a humanized yeast model, lipid phosphatase activity for this variant was functionally neutral (Mighell TL et al. Am. J. Hum. Genet. 2018 05;102:943-955). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
