Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_015443.4(KANSL1):c.767C>T (p.Ser256Phe), citing LabCorp Variant Classification Summary - May 2015: Variant summary: KANSL1 c.767C>T (p.Ser256Phe) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant allele was found at a frequency of 4.1e-05 in 1614174 control chromosomes, predominantly at a frequency of 0.00091 within the East Asian subpopulation in the gnomAD database. c.767C>T has been reported in the literature in a family affected with developmental dysplasia of the hip (Xu_2022). This report however, does not provide unequivocal conclusions about association of the variant with Koolen-De Vries Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
