Submitter rationale: This deletion involves multiple exons (NM_001162501.2) of the 3' portion of TNRC6B (OMIM 610740). Haploinsufficiency of TNRC6B (CCID:008025) is associated with autosomal dominant global developmental delay with speech and behavioral abnormalities (GDSBA) (OMIM 619243; Granadillo 2020, Yang 2024). There are no similar copy number losses of this region in the general populations of the Database of Genomic Variants. Thus, this copy number variant (CNV) is classified as pathogenic.
