This is a single-copy gain (three copies) of the chr21:15520684-30322198 region (~14.80 Mb) on cytogenetic band 21q11.2-21.3. Submitter rationale: Heterozygous duplications of 21q21.3 involving APP have been associated with autosomal dominant inherited early-onset Alzheimer disease (ADEOAD; OMIM 104300; CCID:006677; ISCA-46757; Grangeon 2023, Hooli 2012, McNaughton 2012, Wallon 2012). Individuals with similar duplications showed variable phenotypic presentation, even within families (Grangeon 2023, McNaughton 2012). Two similar overlapping duplications of this locus have been identified in affected individuals (Capkova 2014). There are no similar copy number gains of this region in the general populations of the Database of Genomic Variants. Therefore, based on current medical literature and gene content, this copy number variant (CNV) is classified as pathogenic.
