Pathogenic — the classification assigned by Quest Diagnostics Nichols Institute San Juan Capistrano to GRCh37/hg19 8q24.13-24.3(chr8:126446968-146295771)x3, citing ACMG/ClinGen CNV Guidelines, 2019: This 8q24.13q24.3 duplication involves at least 128 protein-coding genes. Terminal 8q duplications have been reported in individuals with various phenotypes (Bonaglia 2005, Chen 2024, Firth 2009, Hilger 2013, Mergener 2024, Sharma 2013, Sun 2021, Touhami 2023, Yue 2021). There are no similar copy number gains of this region in the general populations of the Database of Genomic Variants. Therefore, this duplication is classified as pathogenic.
