This is a single-copy gain (three copies) of the chr6:142220207-144502854 region (~2.28 Mb) on cytogenetic band 6q24.1-24.2. Submitter rationale: This duplication involves at least 15 protein-coding genes and fully overlaps the 6q24 region (ISCA-37442). Heterozygous duplications of the 6q24 region including PLAGL1 are associated with 6q24-related transient neonatal diabetes mellitus (6q24-TNDM; Temple 2018). Due to imprinting effects of genes in this region, clinical presentation may vary depending on the parental origin of the duplication (Temple 2002). There are no similar copy number gains of this region in the general populations of the Database of Genomic Variants.
