NM_000548.5(TSC2):c.3030G>C (p.Gln1010His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q1010H variant (also known as c.3030G>C), located in coding exon 26 of the TSC2 gene, results from a G to C substitution at nucleotide position 3030. The glutamine at codon 1010 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
