Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005732.4(RAD50):c.3065T>C (p.Leu1022Pro), citing Ambry Variant Classification Scheme 2023: The p.L1022P variant (also known as c.3065T>C), located in coding exon 20 of the RAD50 gene, results from a T to C substitution at nucleotide position 3065. The leucine at codon 1022 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
