NM_000264.5(PTCH1):c.1385G>A (p.Cys462Tyr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C462Y variant (also known as c.1385G>A), located in coding exon 10 of the PTCH1 gene, results from a G to A substitution at nucleotide position 1385. The cysteine at codon 462 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
