This variant lies in the TTN gene (transcript NM_001267550.2) at coding-DNA position 59474, where G is replaced by C; at the protein level this means replaces arginine at residue 19825 with threonine — a missense variant. Submitter rationale: Variant summary: TTN c.51770G>C (p.Arg17257Thr) results in a non-conservative amino acid change located in the A-band domain of the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 6.4e-05 in 248454 control chromosomes (gnomAD). This frequency is not significantly higher than expected for a pathogenic variant in TTN causing Cardiomyopathy (6.4e-05 vs 0.00063), allowing no conclusion about variant significance. To our knowledge, no occurrence of c.51770G>C in individuals affected with Cardiomyopathy and no experimental evidence demonstrating its impact on protein function have been reported.
