Single allele was classified as Pathogenic by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.835C>T (p.R279*) alteration, located in coding exon 8 of the GIGYF1 gene, consists of a C to T substitution at nucleotide position 835. This changes the amino acid from an arginine (R) to a stop codon at amino acid position 279. This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This pathogenic variant has been reported in one individual diagnosed with autism (Chen, 2022). Based on the available evidence, this alteration is classified as pathogenic.
