This variant lies in the POLE gene (transcript NM_006231.4) at coding-DNA position 2141 through coding-DNA position 2142, inserting TGTT; at the protein level this means shifts the reading frame starting at glutamic acid residue 714, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.2141_2142insTGTT variant, located in coding exon 19 of the POLE gene, results from an insertion of 4 nucleotides at position 2141, causing a translational frameshift with a predicted alternate stop codon (p.E714Dfs*78). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay.
