Uncertain significance — the classification assigned by Ambry Genetics to NM_001130009.3(GEN1):c.1325A>C (p.Glu442Ala), citing Ambry Variant Classification Scheme 2023: The p.E442A variant (also known as c.1325A>C), located in coding exon 12 of the GEN1 gene, results from an A to C substitution at nucleotide position 1325. The glutamic acid at codon 442 is replaced by alanine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
