Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002691.4(POLD1):c.3271G>C (p.Asp1091His), citing Ambry Variant Classification Scheme 2023: The p.D1091H variant (also known as c.3271G>C), located in coding exon 26 of the POLD1 gene, results from a G to C substitution at nucleotide position 3271. The aspartic acid at codon 1091 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
