This variant lies in the TTN gene (transcript NM_001267550.2) at 5 bases into the intron immediately after coding-DNA position 2370, where G is replaced by C. Submitter rationale: This sequence change falls in intron 14 of the TTN gene. It does not directly change the encoded amino acid sequence of the TTN protein, but it affects a nucleotide within the consensus splice site of the intron. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a TTN-related disease. Nucleotide substitutions within the consensus splice site are relatively common causes of aberrant splicing (PMID: 17576681, 9536098).
