This variant lies in the TTN gene (transcript NM_001267550.2) at the canonical splice donor site of the intron immediately after coding-DNA position 96904, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The c.69709+1G>A intronic variant results from a G to A substitution one nucleotide after coding exon 174 of the TTN gene. Exon 174 is located in the A-band region of the N2-B isoform of the titin protein and is constitutively expressed in TTN transcripts (percent spliced in or PSI 100%). This alteration disrupts the canonical splice site and is expected to cause aberrant splicing.
