Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003924.4(PHOX2B):c.280G>C (p.Glu94Gln), citing Ambry Variant Classification Scheme 2023: The p.E94Q variant (also known as c.280G>C), located in coding exon 2 of the PHOX2B gene, results from a G to C substitution at nucleotide position 280. The glutamic acid at codon 94 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
