Uncertain significance for Autosomal recessive distal spinal muscular atrophy 1; Charcot-Marie-Tooth disease axonal type 2S — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002180.3(IGHMBP2):c.1208A>G (p.Gln403Arg), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the IGHMBP2 gene (transcript NM_002180.3) at coding-DNA position 1208, where A is replaced by G; at the protein level this means replaces glutamine at residue 403 with arginine — a missense variant. Submitter rationale: In summary, this variant is a novel missense change with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a IGHMBP2-related disease. This sequence change replaces glutamine with arginine at codon 403 of the IGHMBP2 protein (p.Gln403Arg). The glutamine residue is highly conserved and there is a small physicochemical difference between glutamine and arginine.
