Uncertain significance for Arrhythmogenic right ventricular dysplasia 10 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001943.5(DSG2):c.145C>A (p.Arg49Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with serine, which is neutral and polar, at codon 49 of the DSG2 protein (p.Arg49Ser). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with arrhythmogenic right ventricular cardiomyopathy (Invitae). ClinVar contains an entry for this variant (Variation ID: 466331). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This variant disrupts the p.Arg49 amino acid residue in DSG2. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 16773573, 19151369). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing.
