Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000268.4(NF2):c.134A>C (p.Asp45Ala), citing Ambry Variant Classification Scheme 2023: The p.D45A variant (also known as c.134A>C), located in coding exon 2 of the NF2 gene, results from an A to C substitution at nucleotide position 134. The aspartic acid at codon 45 is replaced by alanine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
