NM_000368.5(TSC1):c.3301G>A (p.Glu1101Lys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E1101K variant (also known as c.3301G>A), located in coding exon 21 of the TSC1 gene, results from a G to A substitution at nucleotide position 3301. The glutamic acid at codon 1101 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
