NM_002485.5(NBN):c.386A>G (p.Asn129Ser) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N129S variant (also known as c.386A>G), located in coding exon 4 of the NBN gene, results from an A to G substitution at nucleotide position 386. The asparagine at codon 129 is replaced by serine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
