Submitter rationale: The c.1602_1603delGCinsTG variant (also known as p.E534_P535delinsDA), located in coding exon 13 of the TSC1 gene, results from an in-frame deletion of GC and insertion of TG at nucleotide positions 1602 to 1603. This results in the substitution of the glutamic acid and proline residues for aspartic acid and alanine residues at codon 534 and 535. This amino acid region is not well conserved in available vertebrate species. In addition, this alteration is predicted to be neutral by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Based on the available evidence, the clinical significance of this variant remains unclear.
