Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_004415.4(DSP):c.3912G>C (p.Glu1304Asp), citing Ambry Variant Classification Scheme 2023: The p.E1304D variant (also known as c.3912G>C), located in coding exon 23 of the DSP gene, results from a G to C substitution at nucleotide position 3912. The glutamic acid at codon 1304 is replaced by aspartic acid, an amino acid with highly similar properties. This variant has been detected in an individual with sudden cardiac arrest; however, additional details were limited (Giudicessi JR et al. Int J Cardiol, 2018 Nov;270:214-220). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
