Uncertain significance — the classification assigned by Ambry Genetics to NM_013266.4(CTNNA3):c.605A>G (p.Asp202Gly), citing Ambry Variant Classification Scheme 2023: The p.D202G variant (also known as c.605A>G), located in coding exon 5 of the CTNNA3 gene, results from an A to G substitution at nucleotide position 605. The aspartic acid at codon 202 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
