Uncertain significance — the classification assigned by Ambry Genetics to NM_013266.4(CTNNA3):c.2074G>C (p.Glu692Gln), citing Ambry Variant Classification Scheme 2023: The p.E692Q variant (also known as c.2074G>C), located in coding exon 14 of the CTNNA3 gene, results from a G to C substitution at nucleotide position 2074. The glutamic acid at codon 692 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
