NM_002439.5(MSH3):c.822C>G (p.Cys274Trp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C274W variant (also known as c.822C>G), located in coding exon 5 of the MSH3 gene, results from a C to G substitution at nucleotide position 822. The cysteine at codon 274 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
