NM_012073.5(CCT5):c.895C>G (p.Leu299Val) was classified as Uncertain significance for Hereditary sensory and autonomic neuropathy with spastic paraplegia by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt CCT5 protein function. ClinVar contains an entry for this variant (Variation ID: 465409). This variant has not been reported in the literature in individuals affected with CCT5-related conditions. This variant is present in population databases (rs141509761, gnomAD 0.1%), and has an allele count higher than expected for a pathogenic variant. This sequence change replaces leucine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 299 of the CCT5 protein (p.Leu299Val). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
