Uncertain significance — the classification assigned by Ambry Genetics to NM_016507.4(CDK12):c.4112T>G (p.Val1371Gly), citing Ambry Variant Classification Scheme 2023: The p.V1371G variant (also known as c.4112T>G), located in coding exon 14 of the CDK12 gene, results from a T to G substitution at nucleotide position 4112. The valine at codon 1371 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
