Uncertain significance — the classification assigned by Ambry Genetics to NM_016507.4(CDK12):c.1915G>C (p.Asp639His), citing Ambry Variant Classification Scheme 2023: The p.D639H variant (also known as c.1915G>C), located in coding exon 2 of the CDK12 gene, results from a G to C substitution at nucleotide position 1915. The aspartic acid at codon 639 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
