Uncertain significance — the classification assigned by Ambry Genetics to NM_016507.4(CDK12):c.679T>C (p.Trp227Arg), citing Ambry Variant Classification Scheme 2023: The p.W227R variant (also known as c.679T>C), located in coding exon 1 of the CDK12 gene, results from a T to C substitution at nucleotide position 679. The tryptophan at codon 227 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
