Uncertain significance for Cystic fibrosis — the classification assigned by Ambry Genetics to NM_000492.4(CFTR):c.4419A>C (p.Glu1473Asp), citing Ambry Variant Classification Scheme 2023: The p.E1473D variant (also known as c.4419A>C), located in coding exon 27 of the CFTR gene, results from an A to C substitution at nucleotide position 4419. The glutamic acid at codon 1473 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
